Development of Chromatographic Techniques to Separate and Concentrate pathogenic Micro-organisms from Food - FS1318

The main objective of this research will be to develop straightforward , low cost chromatographic techniques for the separation and concentration of pathogenic bacteria from food suspensions and enrichment broths.
1. To assess the cell surface characteristics in terms of charge and hydrophobicity of the food pathogens Salmonella, E. coli and Listeria. The assessment will take the form of a literature review, consultation with experts and laboratory studies as appropriate ( CFDRA). Surface charge will be determined using electrophoretic mobility and hydrophobicity from retention to hydrophobic gel media. 2. From data obtain in (1) and in consultation with manufacturers, select the most appropriate chromatographic media. 3. To define the sample preparation techniques compatible with the chromatographic separation of bacteria from food debris. Approaches to be investigated include homogenisation and filtration, enzymatic degradation and bacterial precipitation with centrifugation (will consult with LFRA, IFRR and CSL Torry). 4. To quantitatively assess chromatographic media to (a) separate bacteria from food debris and (b) to concentrate bacteria with, if feasible, a broad specificity from pre-enrichment broths. 5. TO establish optimun formats for the chromatographic spearations defined under (4). Column systems, batch treatment and the Pharmacia StreamlineTM expanded bed adsorption technique will be evaluated.
