Method development for the quantitation of equine DNA and feasibility of establishing objective comparisons between measurement expression units (DNA/DNA compared to w/w tissue) - FA0135

Description
Food authenticity and food fraud are becoming increasingly problematic owing to pressures on food production and the current climate of financial constraint. The recent findings of horse DNA present in beef burgers sold in a UK supermarket chain has highlighted the need to provide support for rapid and reliable appraisal of the meat supply chain by developing standardised approaches for the detection and quantitation of different meat products. Samples screened as being suspect in nature need to be subjected to proper testing to fully quantitate the amount of horse DNA present. Measurement uncertainty and accuracy need to be ascribed to real-time PCR approaches (both the original results provided by Ireland and any further approaches), and guidance given on expression units between w/w tissue samples and DNA measurements. This will allow an informed decision to be made based on objective scientific evidence.
Objective
The methodology is split into two main aims:
1) Method development for the quantitation of equine DNA present in processed food samples
2) The feasibility of establishing objective comparisons between DNA/DNA and w/w tissue measurements

These aims will address the issue by establishing accuracy associated with quantitative approaches, and providing guidance on the levels of equivalence between expression units.
